No life-threatening injuries were reported, and at least three people were spotted getting into an ambulance.
The bus also was carrying three DOC staffers.
All inmates and staff were escorted off the bus, the DOC said. Some prisoners at the scene were spotted standing and handcuffed.
The prisoners and staffers were taken to an area hospital following the crash.
The bus was bound for Ulster Correctional Facility, a medium-security prison. The prisoners were being transported there after being held in New York City for court appearances.
The bus apparently rolled over after being cut off on the Thruway. A spokesman told the Associated Press there was no escape attempt.
